The creature has a GUID. You can't have a spawned creature without it not having GUID, there will be a server error if that ever happened. Not sure how to obtain a GUID through SAI, but you can.

What 21 creatures are you talking about? Are you referring to the dungeon not the raid? Or are you talking about the raid bosses all together? I can see that being over 20. Anyway, I'm not 100% sure, but regardless, you can do most of this in SAI. If you aren't SAI friendly I would suggest using C++.
It's all just a simple spawn and move:

boss->GetMotionMaster()->MovePoint(1, x, y, z); // Set x, y, z to correct coordinates
